BRIEF RESUME OF THE INTENDED WORK





    1. Need for the study:

Stress plays an important role in the development of various affective disorders and responsible for various disease like hypertension, diabetes, cardiovascular disorders and effect on reproductive functions1. The reproductive functions changes by life style related factors and with stress involve reduced fertility then impaired hormonal secretions, reduced viability of offspring and related sexual behavior2. And reduced sperm concentration semen quality3. During the past decades several reports have suggested a time and stress related decline in sperm quality4. A meta analysis of 63 studies mainly from USA and from Europe indicated almost 50% reduction from 113x106/ml to 65x106 /ml in the mean semen concentration, during the period 1940 to 19505.

It has been shown that there is a close co- relation between the sperm concentration and fertility potential of male6. In the western society as many as 7.5% of all male factors in at least 25% of such cases. Thus, failure of reproductive organ is encountered by a significant proportion of man and represents significant health burden since reproduction is one of the most important aspect of life of majority of individuals. The present available drugs in the market for the treatment of erectile dysfunction, sildanafil citrate and tadafil. These drugs have severe cardiovascular hypotension, hypersensitivity, myalgia, abnormalities and prolonged causes of infertility7. Only few drugs in the market, which overcomes from the impotency, infertility in male without any side effects.

Ancient people of India in Indian system of medicine has quoted several herbal products to overcome this conditions, on this basis and literature available from traditional claim of Listea glutinosa for its applicability as a aphrodisiac and in infertility, but literature review suggests that no scientific work has been carried out till date. Hence this plant has been selected for the studies. So these research studies helps in overcoming of impotency and reproductive associated problems with respect to environmental changes






6.2 Review of literature:-

Plant Profile:

Title of plant : Litsea glutinosa

Family : Lauraceae

Synonyms : Maidalatki, vasa. Garbijaur, kurkuchita

Parts used : Bark

Chemical constituents: Actinodaphnine, B- sitosterol, boldine, norboldine, laurotetanine,N- Methyllaurotetanine, N- methylactidaphnine, quercetine, kaempferol-3- glucoside, quercetin, kaempferol -7- glucoside, pelargonidine -5- glucoside, aminoacids, mono and sesquiterpenes, sebiferine, litsiferine8, 9.

Medicinal action and uses: According to the treatise on Indian medicinal plants oil from berries is used in rheumatism. Bark is demulcent, aphrodisiac and anodyne, used in the treatment of diarrhea and dysentery sprains rheumatic gouty joints, acts as local antidote to bites of venomouse animals. Leaves are mucilaginous, employed in poultices for wounds and bruises. Decoction of roots used as an emmenagogue. Oil from berries is having anti-rheumatic activity8, 9.

Pharmacological investigation shows this plant has shown scientifically antibacterial 10, cytotoxic activity11.



6.3Objectives of the study:

The botanist in ayurvedic college in our college campus was authenticated bark was collected and extracted using non-polar and polar solvents. The extract used for the present study, with the following objectives:



  1. In vivo evaluation of aphrodisiac activity of Litsea glutinosa extract

  2. In vitro study on effect of extract on various parameters of semen

  3. To observe fertility activity on sterile male rats.

METHODS AND MATERIALS:

7.1 Source of data: For the evaluation of above said activity animals will be subdivided in to following

groups containing six animal in each group


  1. Effect of vehicle on sexual behavior.

  2. Effect of low dose of extract on sexual behavior.

  3. Effect of moderate dose of extract on sexual behavior.

  4. Effect of high dose of extract on sexual behavior.

  5. Effect vehicle normal animal group

  6. Effect of stress on sexual activity

  7. Effect of stress + standard drug on sexual activity

  8. Effect of low dose of extract + stress on sexual activity.

  9. Effect of moderate dose of extract + stress on sexual activity.

  10. Effect of high dose of extract + stress on sexual activity

7.2 Materials :

Plant material : Litsea glutinosa

Animals : Male Wister albino rats

Chemicals : All chemicals to be used in the present study are AR grade

Instruments : Spectrophotometer, triangular microscope, refrigeration centrifuge,

haemocytometer, animal balance etc.

7.3 Methods:

Preparation of Litsea glutinosa extract: The bark authenticated and collected in the ideal collections in the air dried under the shade and powdered to be a fine powder of uniform size by passing through the sieve number 40. The powder collected will be extracted with Petroleum ether, chloroform, ethanol etc. the percent yield will be calculated and dose will be selected based on acute toxicity study12. It will be administered after reconstitution in normal saline





Aphrodisiac activity: Following method with or without modification13 will carry out study. Male rats randomly divided into three groups and these groups will be orally treated with different concentration of extracts. These rats will be individually placed in cages 3 h following the administration and will be given 10 min. adaptation periods. A female that will be brought into oestrus place in cage. After ejaculation or 15 min, after pairing mount frequency, mount latency, ejaculatory latency, % mounted, % ejaculated, copulatory efficiency, intercopulatory interval etc. will be calculated.

In vitro evaluation: In this method after scarifying the animals the sperm motility, sperm count, weight of accessory organs, fructose content, and abnormal sperms will be compared with standard and % will be calculated14.

Anti infertility activity: The male infertility will be induced by following the earlier study, prolonged immobilization (stress) cause a significant delay in testicular maturation and decrease in spermatid production and in sperm density, thus the animals will be exposed regularly for a period of 30 days by immobilization stress for 6 hr in day during light period. The animals with decreased sexual activity will be selected for a evaluation of effect of Litsea glutinosa 15.

7.4 Does the study require any investigation or to be conducted on patients or other humans/animal? If so please describe briefly

Yes, the above study requires to be carried out in male and female albino rats for aphrodisiac and male infertile characters. The in vitro and in vivo evaluation of the effect of Litsea glutinosa requires the use

of rats and also sacrifice to study the sperm characters. This will be done in accordance procedure reported in literature.

7.5 Has ethical clearance been obtained from your institution for performing various tests on animals?

Yes, the study is cleared from institutional animals ethics committee and the copy is enclosed with the protocol.
