摘要】 仙草﹙Mesona procumbens Hemsl.﹚為唇形科仙草屬一年生草本植物,廣泛分布在台灣及中國。在台灣被製作成青草茶和仙草凍,長久被應用在傳統民間用藥。本研究中,仙草全草用甲醇浸泡,其甲醇萃取物經由分配萃取法得到乙酸乙酯層、正丁醇層及水層。在初期的細胞毒殺及抗氧化活性試驗中,乙酸乙酯層及正丁醇層皆有明顯的效果。乙酸乙酯層進一步以管柱層析、高效能液相層析﹙hplc﹚與再結晶方法進行分離純化,經由光譜分析及文獻比對,共鑑定出五十二個化合物,包括十九個三萜類、十個芳香族類、三個生育醇、十二個固醇類、三個雙萜類、一個類黃酮類、一個生物鹼類、一個木酚素類、一個香豆素類及一個其他類之化

摘要

仙草﹙Mesona procumbens Hemsl.﹚為唇形科仙草屬一年生草本植物,廣泛分布在台灣及中國。在台灣被製作成青草茶和仙草凍,長久被應用在傳統民間用藥。本研究中,仙草全草用甲醇浸泡,其甲醇萃取物經由分配萃取法得到乙酸乙酯層、正丁醇層及水層。在初期的細胞毒殺及抗氧化活性試驗中,乙酸乙酯層及正丁醇層皆有明顯的效果。乙酸乙酯層進一步以管柱層析、高效能液相層析﹙HPLC﹚與再結晶方法進行分離純化,經由光譜分析及文獻比對,共鑑定出五十二個化合物,包括十九個三萜類、十個芳香族類、三個生育醇、十二個固醇類、三個雙萜類、一個類黃酮類、一個生物鹼類、一個木酚素類、一個香豆素類及一個其他類之化合物。其中,11α-hydroperoxyurs-12-en-3-one (T12)、22α-hydroxytaraxast-20-en-3-one (T14) 、 20α,21α-epoxytaraxastan-3-one (T15) 為新化合物。

抗氧化試驗方面,化合物ethyl 3,4-dihydroxybenzoate (A5)、sinapic aldehyde (A6)、ethyl 3-(3,4-dihydroxyphenyl)prop-2-enoate (A8)、vinyl caffeate (A9) 與pinoresinol (L1)具有顯著的清除DPPH自由基之能力,其EC50值分別為30.2、69.0、80.0、40.6與48.0 μM。

Abstract



Mesona procumbens Hemsl. (Labiatae), commonly known as hsian-tsao, is an annual herb and widely distributed in Taiwan and China. It is a popular herbal medicine and consumed both as an herbal drink and a jelly-type dessert in Taiwan. In this study, the air-dried whole plant of M. procumbens was extracted with methanol (MeOH). The MeOH extract was partitioned sequentially by ethyl acetate (EtOAc), n-butanol (n-BuOH), and water, then, we collected EA, n-BuOH, and water layers respectively. In the preliminary cytotoxic and antioxidant assay, the EtOAc layer and n-BuOH layer exhibited significant cytotoxic and antioxidant activities. The EtOAc layer was further isolated and purified by column chromatography, high performance liquid chromatography (HPLC), and recrystallization. Fifty-two compounds were obtained through above steps. These compounds are classified into nineteen triterpenoids, ten aromatics, three tocopherol, twelve steroids, three diterpenoids, one flavonoid, one alkaloid, one lignan, one coumarin, and one other type compound. Among these, 11α-hydroperoxyurs-12-en-3-one (T12), 22α-hydroxytaraxast-20-en-3-one (T14), and 20α,21α-epoxytaraxastan-3-one (T15) were isolated from the natural source for the first time. Their structures were determined by spectral analysis and chemical conversion methods. Ethyl 3,4-dihydroxybenzoate (A5), sinapic aldehyde (A6), ethyl 3-(3,4-dihydroxyphenyl)prop-2-enoate (A8), vinyl caffeate (A9), and pinoresinol (L1) showed DPPH free radical scavenging activities with EC50 values of 30.2, 69.0, 80.0, 40.6, and 48.0 μM, respectively.
