5. TITLE OF DISSERTATION:

PREPARATION, PHYSICO

CHEMICAL ANALYSIS AND ANTIMICROBIAL STUDY OF GODANTI BHASMA .

6. BRIEF RESUME OF THE INTENDED WORK :-

6.1 NEED FOR STUDY :

Godanti(Gypsum-CaSO4 7H2O) is well known therapeutic Rasa dravya and it is commonly used in the form of bhasma. Different methods are mentioned in the preparation of Godanti bhasma to get potent and therapeutical properties. It is indicated in Shwetapradar, Shwasa, Kasa, Jwara Kshya etc.

Shwetapradar is one of the most commonest Gynecological problem occurring mainly due to the poor nutrition and unhygienic condition. There are many single drugs and formulations prescribed for Shwetapradar in classics. In which Godanti bhasma is clearly indicated in Shwetapradar. So that this study is selected to evaluate the efficacy of Godanti bhasma in terms of anti microbial activity in Shweta pradar.

Considering above view my work will be carried out based on the following steps:

  1. Idnentification and collection of raw material according to its grahya laxanas and by modern methods.

  2. Physico-chemical analysis of ashodita and shodita Godanti.

  3. Prepartion of Godanti Bhasma and its physico chemical anlysis.

  4. .In vitro evaluation of its bacteriostatic, bacteriocidal and antifungal activity responsible for Shwetapradar.

6.2 REVIEW OF LITERATURE :-
The grahya Godanti possess the colour as that of shweta (Sharada kalina chandra), patrachita, sumasrunam, nirmalam and diptaprabham.1 Godanti is having sheeta virya. It acts as Jwargna, Kshaya, kasa, panduroghara, urakshata and it is specially indicated for balashosh and Shwetapradara2.

Various methods of shodhana have been explained for Godanti(Gypsum CaSO4) in classical texts are,

I METHOD. Godanti is subjected to swedana in a dola yantra filled with Nimbu(Citrus aurantifolia christm) swarasa for ardha yama.3

II METHOD: Godanti is subjected to swedana in a dola yantra filled with Dronpuspi(Leucas cephalotes Spring). Swarasa for ardha yama.3

III METHOD: It’s Shodhana can be done by washing it with hot water4.

PREPARATION OF GODANTI BHASMA:

I METHOD: The bhavana of Nimba patra(Azadirachta Indica A.Juss) swarasa is given to the ShuddhaGodanti churna for one day, prepare chakrikas and subject to gajaputa.By this method Shweta varna bhasma is obtained5.

II METHOD: To shuddha Godanti churna bhavana is given with Kumari(Aloe vera.Tourn ex.Linn) swarasa for one day, prepare chakrikas and subject to gajaputa. By this method Shweta varna bhasma is obtained.6

III METHOD: Small pieces of shuddha godanti is kept inside the sharava samputa and subjected to gajaputa. In this method its Shweta varna bhasma is obtained7.

Previous work done on Godanti bhasma w.r.t Shweta pradara

1.1994-Raman. Trivendram A study of Godanti bhsma w.s.r. to its therapeutic effect in Shweta pradar.

2.1995- Rajam. R. Trivendram A study of Godanti bhasma w.s.r. to its therapeutic effect in Shweta pradar.

    1. OBJECTIVES OF THE STUDY :

  1. Identification and collection of raw materials according to classical and modern

method.

  1. Physico-chemical analysis of raw materials before and after shodhana.

  2. Physico chemical analysis of Godanti bhasma according to G.M.P adopting S O P

  3. Invitro evaluation of its bacteriostatic, bacteriocidal and antifungal action rersponsible

for shweta pradara..
7. MATERIALS AND METHODS :-

7.1 Source of Data :-

  1. Samples of Godanti(Gypsum CaSO4) will be collected from the market and get authenticated by the experts in the Rasa shatra subject.

  2. Samples of Nimbu, Nimba patra will be collected from the garden of Shri B.M.Kankanwadi Ayurvediya Mahavidyalaya and get aunthenticated by the experts in the Dravya guna subject.

  3. The Literary data will be collected from the Library of Shri B.M.K. Ayurveda Mahavidyalaya, internet and Magazines.

  4. Physico-chemical analytical data will be collected from well recognised Laboratory.

  1. The antimicrobial study will be conducted in the well recognized microbiological Laboratory..

7.2 METHODOLOGY ;

  1. The sample of Godanti which is having grahya laxana’s like shweta varna (as sharada kalina chandra), patrachita, sumasrana, nirmalam, diptaprabham1 will be collected.

2. Shodhana of Godanti :-

Shodhana of Godanti will be carried out by giving swedana in Nimbu (Citrus aurantifolia christm) swarasa by using Dola yantra.for ardha yama.3.

  1. Preparation of Godanti Bhasma.

To Shuddha Godanti churna Nimba patra(Azadirachta Indica A.Juss) Swarasa bhavana is given for one day and it is made into chakrikas. Then after drying kept in sharava samputa and subjected to gajaputa. After swanga sheeta it is taken out.By this method Godanti bhasma is prepared5.
The Physico chemical changes will be observed in stepwise manner before and after shodhana of Godanti and after preparation of Godanti bhasma w.r.t. organoleptic characters like texture, colour, taste form and odour and its pH value and same will be noted.
Physico chemical analysis of Ashudha, shodhita and Godanti bhasma is carried as follows:

  • Physical Method.

    • Weight. Melting Point.

    • Structure. Boiling Point.

    • Colour. Solubility.

    • Odour. Consistency.

    • Taste. Luster.

    • pH value.

Chemical analysis-

Quantitative Method:

-Estimation of percentage of Calcium..

    • Estimation of percentage of Sulphate..

    • Loss on drying.- finding the moisture content drugs.

    • Ash value - for analyzing percentage of inorganic constituents of Bhasma.

    • Acid insoluble ash.- for assessing any silica or mud particle in drugs.

  • Qualitative Method: -

    • Trace elements analysis.

  • Instrumental Method: -

    • X R D

    • A A S

Antimicrobial study :-

Methodology - Microbroth Inhibition Concentration(M I C) .

Media-Mueller Hinton Broath.

Total 4 micro-organisms and one fungus will be included to assess the anti-microbial efficacy of the compound.

Two gram Positive bacteria – Staphylococcus aureus and Streptococcus agalactiae

Two gram Negative bacteria - Escherichia coli and Klebsiella.

One fugas- Candida albicans.will be included to assess the anti microbial efficacy of the compound.

The anti-microbial activity will be carried out by Microbroth dilution method. Here serial dilution of the compound will be tested against fixed density of each bacteria. Results will be expressed as Minimum Inhibitory Concentration (microbes). After overnight incubation, specimen from each tube, which shows turbidity, will be inoculated on blood agar plate. The absence of growth will indicate bactericidal activity of particular concentration of the compound against tested bacteria.
7.3 DOES THE STUDY REQUIRE ANY INVESTIGATIONS OR INTERVENTIONS TO BE CONDUCTED ON PATIENTS OR OTHER HUMANS OR ANIMALS

--- Not required

7.4 HAS ETHICAL CLEARANCE BEEN OBTAINED FROM INSTITUTION IN CASE OF 7.3

--- Not required
